MAbs:
3A1, 1G4, 1D6Hybridoma clones have been derived from hybridization of X63-Ag8-653 myeloma cells with spleen cells of Balb/c mice immunized with purified human proinsulin.
Specificity:
MAbs 3A1 and 1G4 are directed to different antigenic determinants. MAb 3A1 reacts with the site of connection of C-peptide and C-end of B-chain of proinsulin molecule.MAbs 1D6 reacts with different epitopes on human proinsulin molecule; both of each are generated by the interactions between insulin and C-peptide parts of prohormone.Kd for MAb 3A1 is 1.1 x 10-8 M.Kd for MAb 1G4 is 2.6 x 10-7 M.Kd for MAb 1D6 is >3 x 10-8 M.MAbs 3A1, 1G4, 1D6 do not cross-react with human, bovine and porcine insulin,bovine and porcine proinsulin, and human C-peptide.
